Youngner, Julius S. (University of Pittsburgh, Pittsburgh, Pa.), and Marion E. Kelly. Inhibition by exogenous interferon of replication of poliovirus ribonucleic acid in chick brain. J. Bacteriol. 90:443-445. 1965.-The replication of poliovirus was studied after the intracerebral inoculation of infectious ribonucleic acid (RNA) into the brains of 2-day-old chicks; these animals are not susceptible to intact virus. Single-cycle replication of virus, which reached a peak in about 12 hr, was completely inhibited by prior intraperitoneal injection of interferon prepared from the allantoic fluid of chick embryos infected with influenza virus. A single dose of as little as 500 units of interferon, measured by a plaque-reduction method, completely suppressed viral replication when injected 24 hr prior to infectious RNA. This system provides a model for the study of the protection of target organs by passively transferred interferon injected at a distance.

Viral events necessary for the induction of interferon in chick embryo cells.
Temperature-sensitive mutants of Sindbis virus were employed to investigate the nature of the viral event(s) which induces chick-embryo cells to produce interferon. Chick embryo cells induced by the parental heat-resistant strain of Sindbis virus produced essentially equal amounts of interferon at 29 and 42 C. An RNA(-) and three RNA(+) strains [temperature-sensitive mutants unable (RNA(-)) and...
متن کاملEffect of Activation and Inhibition of Cellular PKR on Coxsackievirus B3 Replication
The ds-RNA activated protein kinase (PKR) is a serine-threonine kinase with MW of 68 KDa. It belongs to a family of kinases that control one of the translational initiation factors, eIF2. PKR is produced at high level in response to viral infection. This protein by phosphorylating eIF2 inhibits cellular protein synthesis. In this study, the effect of gamma interferon (IFN-γ), an activator, and ...
متن کاملInhibition of Sindbis virus replication in HeLa cells by poliovirus.
Concomitant infection of HeLa cells with poliovirus and Sindbis (SB) virus allowed replication of poliovirus only. The poliovirus-induced interference with the replication of SB virus was dependent partially on the multiplicity of SB virus used for infection. The observed interference was not sensitive to guanidine. Translation, but not replication of poliovirus, appears to be needed for the re...
متن کاملEffect of inactivation by hydroxylamine on early functions of poliovirus.
Evidence is presented that poliovirus particles with a single lethal hit by hydroxylamine do not induce in host cells either inhibition of cellular protein synthesis or viral ribonucleic acid (RNA) replication. The RNA of these viruses is not replicated even if the cells are simultaneously infected with both active and inactivated viruses. The damaged viral RNA seems to have lost both its templ...
متن کاملCellular SRC kinases and dsRNA dependent protein kinase (PKR) play key role in intracellular viral (CVB3) replication
SRC kinases and PKR are intracellular protein kinases, which play key roles in intracellular viral replication. In this research, the effect of SRC kinase inhibition and PKR activation and inhibition on replication of coxsakievirus (CVB3), an entrovirus of the family picornaviridae – causative agents of fatal myocarditis, was studied. Vero and Hela cells were cultured and infected with CVB3 in ...
